چکیده
The primary virulence of soft rot pectobacteria, including Pectobacterium brasiliense, is mainly determined by the massive production plant cell wall-degrading enzymes (PCWDEs), which promote tissue maceration in many crops. antibiotic treatment to treat bacterial causes environmental problems and potentially affects resistance. Antibiotic resistance driving interest antimicrobial treatments, no organism has been reported have acquired honey. However, use honey as a therapeutic agent for pathogens rarely studied. Therefore, this study was undertaken determine vitro effect Manuka against P. brasiliense at phenotypic genotypic levels. A sublethal concentration growth inhibition assay broth medium containing different concentrations macerating performed on orchid leaves, activities PCWDEs were examined plate assays. expression PCWDE-associated genes investigated using semi-quantitative reverse transcription-polymerase chain reaction analysis. exposure significantly decreased ability synthesis PCWDEs, i.e., pectate lyase, polygalacturonase, protease. Moreover, five PCWDEs-encoding genes, such pelA, pelB, pelC, pehA, prtW, had lower levels after compared with recA internal standard. associated PCWDEs. reduced suppressing PCWDE genes.

The Rcs phosphorelay modulates the expression of plant cell wall degrading enzymes and virulence in Pectobacterium carotovorum ssp. carotovorum.
Production of plant cell wall degrading enzymes, the major virulence factors of soft-rot Pectobacterium species, is controlled by many regulatory factors. Pectobacterium carotovorum ssp. carotovorum SCC3193 encodes an Rcs phosphorelay system that involves two sensor kinases, RcsC(Pcc) and RcsD(Pcc), and a response regulator RcsB(Pcc) as key components of this system, and an additional small lip...
متن کاملDiversity of Beetle Genes Encoding Novel Plant Cell Wall Degrading Enzymes
Plant cell walls are a heterogeneous mixture of polysaccharides and proteins that require a range of different enzymes to degrade them. Plant cell walls are also the primary source of cellulose, the most abundant and useful biopolymer on the planet. Plant cell wall degrading enzymes (PCWDEs) are therefore important in a wide range of biotechnological processes from the production of biofuels an...
متن کاملManuka honey reduces the motility of Pseudomonas aeruginosa by suppression of flagella-associated genes.
OBJECTIVES Manuka honey is a broad-spectrum antimicrobial agent that seems to affect different bacteria in many different ways. It has been shown to be bactericidal against Pseudomonas aeruginosa by destabilizing the cell wall, but we aimed to investigate whether there were further intracellular target sites. METHODS In this study inhibitory effects of manuka honey on P. aeruginosa were inves...
متن کاملArsenal of plant cell wall degrading enzymes reflects host preference among plant pathogenic fungi
BACKGROUND The discovery and development of novel plant cell wall degrading enzymes is a key step towards more efficient depolymerization of polysaccharides to fermentable sugars for the production of liquid transportation biofuels and other bioproducts. The industrial fungus Trichoderma reesei is known to be highly cellulolytic and is a major industrial microbial source for commercial cellulas...
متن کاملExamination of the Role of Fungal Cell Wall- Degrading Enzymes in Plant Fungal Resistance
Transgenic technology has demonstrated its usefulness in weed and insect control, but this technology has yet to expand much beyond these limited, special case scenarios. Along with stringent regulations, the primary limitation preventing expansion into new areas is finding efficacious genes. Fungal pathogens are particularly difficult to control in this regard as these organisms employ many of...
